Product Description
Most information about the nature of living cells is discovered through light microscope observation. The image seen through a microscope is not a magnified point-by-point representation of the object, but is an abstract representation of the object that must be interpreted. This interpretation is influenced not only by the actual structure of the object, but the physical characteristics of light and the microscope components, the preparation of the sample, and the microscopist's background. In order to interpret the image correctly and fully, the microscopist must understand the interaction of light with the sample.

The purpose of this book is to provide the most comprehensive, easy-to-use, and informative guide on light microscopy. Light and Video Microscopy will prepare the reader for accurate interpretation of an image and understanding of the living cell. With the presentation of geometrical optics, it will assist the reader in understanding image formation and light movement within the microscope. It also provides an explanation of the basic modes of light microscopy and the components of modern electronic imaging systems and guides the reader in determining the physicochemical information of living and developing cells which influence interpretation.

* Brings together mathematics, physics, and biology to provide a broad and deep understanding of the light microscope

* Clearly develops all ideas from historical and logical foundations

* Laboratory exercises included to assist the reader with practical applications

* Microscope discussions include: bright field microscope, dark field microscope, oblique illumination,phase-contrast microscope, photomicrography, fluorescence microscope, polarization microscope, interference microscope, differential interference microscope, and modulation contrast microscope

4-0 out of 5 stars A book with useful protocols
This is a book covering the principles of all kinds of microscopes with structure figures. There many good color photos and graphs. The protocols of tissue preparations are very useful, butmaybe duplicate in other books. If you just want to know how to use the microscopes instead of understanding them, it should be a good book for you. It is more like a lab manual collection.

The problem of this book is that it distracts a little bit from microscopes to cell culture and immunohistochemistry. ... Read more

The investigation and manipulation of matter on the atomic scale have been revolutionized by scanning tunneling microscopy and related scanning probe techniques. This book is the first to provide a clear and comprehensive introduction to this subject. Beginning with the theoretical background of scanning tunneling microscopy, the design and instrumentation of practical STM and associated systems are described in detail, including topographic imaging, local tunneling barrier height measurements, tunneling spectroscopy, and local potentiometry. A treatment of the experimental techniques used in scanning force microscopy and other scanning probe techniques rounds out this section. The second part discusses representative applications of these techniques in fields such as condensed matter physics, chemistry, materials science, biology, and nanotechnology, so this book will be extremely valuable to upper-division students and researchers in these areas. ... 5-0 out of 5 stars My opinion about"Scannig Probe Microscopy and Spectroscopy".
This book is one of the principal lecturesthat every research need to understand "Scanning probe apparatus". Its content refer all the different areas of actual interest and the different type of measurements that actually could be make with this type of system. This book is more than a simple introduction in this wonderful area of physics. For all this reasons I thing that is one of the most important books that every research have to read.

3-0 out of 5 stars Poor print quality
The price of this paperback of SPM book is very good compared to that of the hardcover. However, the print quality is poor. In some of the images you barely see the featutres they talk about in the book. I am a SPM user so this is really bothering me. The SPM imagaes in this book are not very informative due to the poor print quality. Maybe the hardcover one has better print quality. ... Read more

Product Description
Since 1995, the noncontact atomic force microscope (NC-AFM) has achieved remarkable progress. Based on nanomechanical methods, the NC-AFM detects the weak attractive force between the tip of a cantilever and a sample surface. This method has the following characteristics: it has true atomic resolution; it can measure atomic force interactions, i.e. it can be used in so-called atomic force spectroscopy (AFS); it can also be used to study insulators; and it can measure mechanical responses such as elastic deformation. This is the first book that deals with all of the emerging NC-AFM issues.

FROM THE REVIEWS:

MATERIALS TODAY"This book gives a comprehensive overview of the state-of-the-art of this dynamic force microscopy technique in 20 chapters, each written by experts in the field. It covers the theoretical basis, as well as applications to semiconducting surfaces, ionic crystals, metal oxides, and organic molecular systems including thin films, polymers, and nucleic acids . . . There are unsolved questions about the mechanisms responsible for atomic resolution but, as this well-written book displays, there has been tremendous progress in basic understanding of the technique and fascinating new applications continue to arise . . . With an increased understanding of NC-AFM, as demonstrated in this book, we are certain to see further progress in the near future." ... Read more

Product Description
Introduction to Electron Microscopy for Biologists is ideal for the scientist who may be considering electron microscopy as a tool to extend molecular, biochemical, or light microscope observations to the next level of structural information, only available by electron microscopy. Each chapter briefly surveys the present state of structural information in a particular area, be it an individual but widely occurring molecule such as actin or collagen, together with the methods for visualization, either as an extracted and purified entity, or in situ within its biological context. Not only is this book an introduction to electron microscopy in general, but it is also useful for those within the field who wish to move to a different area of expertise, for instance an approach based on rapid freezing, rather than more conventional protocols. This should be a first choice reference for any biologist wanting to know 'what does it look like' across the full spectrum of cell and molecular biology of life science.

*Illustrates the advantages of electron microscopy and the protocols required for fine structural visualization of individual molecules upwards, ranging from mammalian tissues to experimental organisms.
*Comprehensive chapters cover both structural information on each area as well as the protocols themselves
*Includes methods covering specific labeling, as well as methods for quantification and three dimensional reconstruction from molecular to tissue levels. ... Read more

2-0 out of 5 stars Not an INTRODUCTION!!
This book claims to be an "Introduction" to electron microscopy, but it is not. Instead, it is a general description of VERY SPECIFIC methods, such as high pressure freezing, electron probe X-ray microanalysis, types of immunolabeling and specific techniques for each cellular organelle. IT DOES NOT CONTAIN ANY GENERAL METHOD OF PRINCIPLE, such as common fixatives, staining methods or embedding resins, nor does it say anything about the physics of the microscope.

If you are really looking for an INTRODUCTION (and a very general one, for that matter) to biological electron microscopy, I would recommend Bozzola's and Russell's Electron Microscopy. It contains general principles of both types of electron microscopes, basic techniques for standard methods, and chapters on more specific methods, like immunocytochemistry, autoradiography and freeze methods. For better and in-depth information about specimen preparation, Hayat's Principles and Techniques of Electron Microscopy is the best book available, although it does not contain any information on the physical principles of the electron micrsocope.

However, if your subject of interest are the physical principles of the electron microscope (and you are a biologist), although a bit old, Sjöstrand's book Electron Microscopy of Cells and Tissues is a very good reference, as well as Wischnitzer's Introduction to Electron Microscopy and Meek's Practical Electron Microscopy for Biologists. ...
