Jiz v roce 1927 spolupracoval s Fermim a stal se asistentem na univerzite v Rime. Od roku 1936 pusobil jako profesor na univerzite v Palermu, ale jiz po dvou letech emigroval do USA, kde nastoupil v ustavu v Berkeley. V Berkeley pracoval nejprve na vyvoji jaderne bomby, ale v roce 1946 se stal na mistni univerzite profesorem. Segre se venoval v nejvetsi mire atomove a jaderne fyzice, zabyval se spontannim stepenim jader. Spolecne s Fermim studoval interakci neutronu s atomovymi jadry a jejich vliv na jaderne reakce. Pri bombardovani molybdenu deuterony vznikl prvni umely prvek - technecium - s protonovym cislem 43, ktery se v prirode nevyskytuje a je pravdepodobne produktem stepeni uranu a plutonia. V roce 1940 byl pri vyrobe astatu s protonovym cislem 33 a v roce 1941 pri vyrobe transuranu plutonia s protonovym cislem 94. Posledni zmineny prvek je vyznamnym stepnym materialem pro jaderne zbrane a palivem pro jaderne elektrarny. Nejvetsim Segreovym objevem byl v roce 1955 objev antiprotonu a od te doby se venoval jiz jen vyzkumu antinukleonu. V roce 1959 obdrzel spolecne s Chamberlainem Nobelovu cenu za objev antiprotonu.
