Ya durante su época de estudiante de Biología y Química en la Universidad de Cambridge, Hodgkin comenzó a interesarse por los fundamentos de la conducción nerviosa, donde ambas materias convergían para explicar uno de los grandes problemas de la fisiología. Inició algunos experimentos bastante sencillos en los nervios de ranas y continuó en la misma línea durante varios años. De forma accidental descubrió que se podían extraer fácilmente las fibras nerviosas individualizadas de un cangrejo de tierra, de modo que resultaban apropiadas para realizar experimentos pese a su reducido tamaño (0,03 mm). Durante el período de 1937 a 1938 Hodgkin pasó varias semanas en los EEUU, donde tuvo ocasión de diseccionar y estudiar las fibras nerviosas del calamar, cuyas dimensiones superaban a las del cangrejo (hasta más de 1 mm de diámetro). En 1938 regresaba a Cambridge, donde empezaba un año después su colaboración con A.F. Huxley, a quien tuvo la fortuna de tener como alumno. La guerra, como a tantos otros científicos de la época, le obligó a interrumpir sus investigaciones sobre el sistema nervioso. Desde 1939 hasta 1945 trabajó en el desarrollo del radar para el Ministerio del Aire. Al terminar la contienda, regresó a Cambridge, primero como conferenciante y más tarde como director adjunto de investigación. Reanudó entonces sus trabajos con el profesor Sir Andrew Huxley, investigando la 'teoría iónica' sobre la transmisión de mensajes al cerebro a través de las células nerviosas.
