Plant Physiology
Overview
Wolf B. Frommer
Physiology studies the function of an organism, encompassing the dynamic processes of growth, metabolism, reproduction and interaction with other organisms. Modern physiology combines a wide spectrum of techniques such as molecular biology, biochemistry, cell biology, genetics and biophysics to obtain insights into the functioning of an organism. Understanding the functioning of a plant organism in its native environment at the whole plant level requires information about the integration of processes, which can only be achieved by physiological research. The availability of the full genome sequences of different organisms including plants, in combination with novel technologies from the areas of genomics, proteomics and metabolomics, provides an enormous resource. Especially the analysis of knock-out mutants and transgenic plants will lead to a quantum leap for this area. The majority of research in our department can be classified as basic research. However, the transfer of results and knowledge could lead to potential biotechnological applications in the fields of crop improvement (yield potential, quality traits) and agrochemistry (crop protection).

The year 1848 not only saw political civilian uprise in Austria but also brought along revolutionary changes for the universities. The new laws concerning university-organisation led to the separation of the flourishing Natural Sciences from the Faculty of Medicine. Franz UNGER, Vienna's first true plant physiologist ("Anatomie und Physiologie der Pflanzen", 1855), graduated with a degree in medical sciences, was a true all-rounder with a profound knowledge in anatomy, physiology, ecology, pathology and paleontology of plants. He was nominated professor for Physiological Botany in 1849, but there was no institute for him to move into. He had to change repeatedly into various places within the university quarter until he resigned in 1866. continue... Page:

Specific questions are (1) what is the molecular mechanism behind vascular strand formation? (2) which genes regulate fiber differentiation? (3) which genes regulate the rate of wood formation and the cellular composition of wood? We focus our research on two model plant species, Arabidopsis and poplar, since considerable genetic resources are available for both species. In Arabidopsis we are using both traditional mutagenesis and reverse genetics to approach gene function, whereas reverse genetics is the preferred choice in poplar. Studies on Plant Physiology and Molecular Biology concentrate in understanding of the structure, function and development of plants at the level of genes, molecules, cells, and a whole plant. The degree program also covers the specific topics of plant biotechnology, stress physiology, and plant/microbe interaction. Research activities of the Laboratory focus on the molecular aspects of plant development and tolerance against environmental stresses, as well as on plant biotechnology. Arabidopsis is a model organism in plant science. The entire genomic DNA sequence of Arabidopsis , now available for scientists, accelerates progress to understanding development and responses of plants to the environment. Another model organism is a cyanobacterium Synechocystis 6803, the genome of which has also been sequenced. The gene chips and proteomic techniques, utilizing the whole sequence information of these model organisms, are currently under development in the Laboratory. In addition, virus-derived vectors are developed for silencing of specific gene functions in the host plants. The largest group in the Laboratory studies the assembly and function of the photosynthetic apparatus as well as the regulation of genes involved in photosynthesis. The most recent research projects in this area are related to the molecular signalling within chloroplasts and between the chloroplasts and the nucleus. Particular interest is focused on the regulation of photosynthesis genes and thylakoid protein phosphorylation by redox signals in chloroplasts. The group uses both plants and cyanobacteria as model organisms and combines molecular biology with expertise in plant physiology and biochemistry.
