Fungal Genetics Research
Yeasts and filamentous fungi are eukaryotic micro-organisms amenable to genetic analysis whcih have been used extensively as model systems for studying aspects of eukaryotic cell biology including growth, division and genetic recombination. Filamentous fungi give us antibiotics that revolutionised medicine, and yeast as a model system provided key insights in understanding cancer through genetic analysis of the cell cycle.
Fungal pathogens such as Candida albicans and Aspergillus fumigatus have recently become major causes of death through infection in immunocompromised patients following heart and lung transplants or anti-cancer chemotherapy.

http://nrs.harvard.edu/urn-3:hul.eresource:funggene Abstract: Fungal Genetics and Biology, formerly known as Experimental Mycology, publishes experimental investigations of fungi and their traditional allies that relate structure and function to growth, reproduction, morphogenesis, and differentiation. This journal especially welcomes studies of gene organization and expression and of developmental processes at the cellular, subcellular, and molecular levels. The journal also includes suitable experimental inquiries into fungal cytology, biochemistry, physiology, genetics, and phylogeny. This journal from Academic Press has online abstracts available from 1993 and full text of articles available online from 1997. Essential Fungal Genetics
COMING SOON To be published in Spring 2002 by Springer-Verlag, New York (currently in production) Essential Fungal Genetics provides the tools for integrating fungal genetics into current teaching at undergraduate/graduate level. It is a user-friendly specialist account, which will complement the major textbooks used in courses on general genetics, general organismal biology, general microbiology and general mycology. Essential Fungal Genetics incorporates all aspects of eukaryote genetics appropriate to fungi, described here in a way that is easily understood and memorable. The book has an instructional ‘how to do it’ tone and will have a practical value for anyone who has a mind to start genetic analysis of any fungus. Not in terms of technical details like laboratory recipes, but more about details of the ideas and concepts underlying basic approaches to fungal genetics so that these are on record and easily accessible. The authors have maintained a readable style of presentation, and provide a range of reference materials at the end of each chapter, favoring recent review articles and websites, so that the interested reader can very quickly penetrate deeper into the literature. David Moore has taught genetics at the University of Manchester since 1967. His research on fungal genetics and development has appeared in over 150 research papers. His books include

Fungal Genetics Lab You can do this lab on the Web or in one of the campus computer labs as announced in class. The lab exercises comprise 5 integrated exercises modified from some of those in 'The Virtual Genetics Lab' section of 'Visual Genetics'. They represent a typical genetic analysis by classical means of a fungus:- 1. first get mutants (in this case auxotrophs)
    2. then classify them for allelism - in this case by biochemical tests
    3. another way to classify them for allelism - in this case by a complementation test
    4. then map the genes by looking at random products of meiosis
    5. another way to map genes and this time also centromeres, by analysis of whole tetrads
(i.e groups of 4 products of one meiosis)
START COMPUTER BASED FUNGAL GENETICS EXERCISE HERE Click here to begin the lab exercises on Fungal Genetics.
Note - after completing the program you need to record and hand in your results. The program allows you to print out a record of your answers. Remember to print them before you close down the program otherwise your work will be lost. If you do not complete the assignment in one sitting - simply print out what you have done and then print another report when you go back to complete the work - then hand in all reports.

VIIIo-1 Nitrogen metabolite signalling and transcript stability in Aspergillus nidulans I. Morozov, MG Jones and MX Caddick, The University of Liverpool, Crown Street, Liverpool, L69 7ZD, UK Nitrogen metabolism in A. nidulans is subject to regulation by the principal GATA transcription factor AREA. One signalling mechanism, which modulates AREA activity, involves regulated degradation of the areA transcript in response to ammonia (Platt et al , EMBO J. 1996 15 2791 – 2801). The specificity of transcript degradation reveals a qualitative response to several further nitrogen sources including Gln and Asn. Both ammonia and Asn need to be converted to Gln via Glu before they are effective as a signal ( Morozov et al Mol Microbiol 2001 42 269 – 277). Recently, we have found that a variety of structural genes and at least one other regulator gene re also subject to the same regulatory mechanism. In the case of areA this mechanism requires a sequence of 218 nucleotides within the 3 untranslated region of the gene. This sequence functions independently and is sufficient to accelerate degradation of the poly(A) tail which in turn leads to rapid transcript degradation (probably via de-capping of the 5
