CENTER NEWS - THURSDAY, JULY 17, 1997 OFF THE TOP Cryo handles cells between harvesting, transplant I n the early days of bone marrow transplantation, researchers aimed to get the cells out of the donor and into the recipient as quickly as possible in hopes they would take. However, in the years since, they've learned there's a lot they can do to the cells while they're "out of body" to improve chances of a successful transplant. For example, donor and recipient need no longer be the same ABO blood type: The red blood cells can be removed from the marrow. Even more exciting are new techniques to remove or enhance immune-reactive cells in order to reduce the risk of graft-versus-host disease or disease relapse after transplantation. At the Center, the handling of cells between harvesting and transplantation is the domain of the Cryobiology Laboratory, three floors below ground level in the Columbia Building and open seven days a week.

Medical Cryobiology Unit The medical cryobiology unit carries out research on basic mammalian cryobiology and applies that knowledge to the development of preservation techniques for cells, tissues and organs of medical interest. Current areas of study include the identification of the factors that, in general, make it more difficult to cryopreserve tissues than cells, and organs than tissues. Specific areas of study include mechanisms and patterns of ice crystallisation in tissues, the role of thermal stress particularly in aqueous glasses, effects on cell adhesion and the use of dielectric heating. The tissues currently studied include corneas, arteries, heart valves, cartilage and bone. The Unit houses the Editorial Office of the journal CRYOBIOLOGY. The research is financed by the UK research councils, the European Community, charities and commercial sources. Further information:

Cryobiology is the branch of biology that deals with the study of life at
low temperaturesgenerally considered to be below freezing. Sometimes,
cryobiology is simply a matter of surviving cold temperatures until times
get better.
Here are some ideas for experiments dealing with cryobiology.
Place different seeds in the freezer overnight. Then plant the seeds and see if they grow. You can make this quantitative by planting many of each kind of seed. Compare the same number with and without the overnight freezer treatment. Is there a difference? Were the seeds affected by the treatment? Place plants at different developmental stages in the freezer overnight, then pull them out and see if they continue to grow. For example, start with a large bean seed. Put the seed in the freezer. Then try the bean when it has taken up water, but the root has not yet emerged (say overnight on a wet paper towel). Now try a seedling with the root emerged, another

Well, Trevor, it would be exciting to be able to do this. As you probably
have learned, many insects survive temperatures below freezing. Many fish
do this also. Even snakes. For example:

The trick is to
prevent the formation of ice crystals, which destroy cells by essentially
tearing them apart. In all instances where this has been studied, the animals have devised some means of preventing ice crystal formation. One way is to not have any water in the cells. This is method used by brine shrimp, which lay eggs that have become dried inside the mother. You can buy brine shrimp eggs (actually they are not eggs but embryos with a few hundred cells) at pet stores. They can be hatched by placing them in dilute salt water. The usual way to prevent ice crystals is with "antifreeze". Insects and fish produce a kind of antifreeze in the form of a sugar solution that either prevents freezing unless much colder than water freezing temperature or by reducing the size of water crystal to where they cause only slight damage. In laboratories people freeze cells by soaking them in a sugar

SUMMARY
Over 60% of whole land on the earth was exposed to sub-zero temperature at least in winter. It is generelly thoght that plants have originated from the climates corresponding to the tropics and the subtropics. They have acquired adaptability for coldness and have expanded almost all land in the earth, except for the polar region at present. Our lab researches to explain the mechanism of adaptation to cold environment and response to environmental stresses in plants by techniques of physiology, biochemistry, molecular biology and so on.
