Nobel Laureates Letter
May 14, 2001 The Honorable C.W. (Bill) Young
Chairman
House Appropriations Committee
H 218 Capitol
Washington, DC 20515: Dear Mr. Chairman: Last year Congress made a bipartisan commitment to strengthen American science and engineering by significantly boosting basic research investments in agencies such as the National Science Foundation, the Department of Energy and the National Institutes of Health. Federal investments in these agencies provide the foundation for long-term economic growth, diagnosis and treatment of disease and improvement in our quality of life. Increasing the federal investment in basic research at the NSF and DOE is particularly important now to help reverse declines in federal spending relative to the U.S. economy over the last decade. As Congress moves forward in its effort to double the budget of the National Institutes of Health, it should recall that today's miraculous advances in biomedical research could never have occurred without significant technological and conceptual contributions from the physical and computational sciences. Before we can reap the full benefits of the human genome project, we will require many further advances in these and other areas. During this year's debate on the FY 2002 budget, the Senate, at the urging of Senators Kit Bond (R-MO) and Barbara Mikulski (D-MD), passed an amendment by unanimous consent that would add $1.44 billion above the presidential request to Function 250, which includes the National Science Foundation, NASA and the Office of Science at DOE. The amendment would allocate an additional $674 million to the NSF and $469 million to the DOE Office of Science. We believe the Senate made the right decision, and we urge you to incorporate these increases into the House Appropriations bills.

Scientist's Nobel Prize-winning research first presented at ORNL conference
from staff reports The idea that eventually won Gunter Blobel the Nobel Prize was first presented in 1971 at an Oak Ridge National Laboratory conference, according to a scientist who was there. John S. Cook, who worked at ORNL at the time and chaired the organizing committee of the Biology Divsion's annual conference, remembered Blobel's presentation in an e-mail to The Oak Ridger. Cook now lives in Falmouth, Mass. Cook wrote that a well-known cell biologist of the day had been invited to the conference. He sent Blobel, then a post-doctoral student, instead. "We were very annoyed to lose a senior scientist and get a junior unknown in his place," wrote Cook. The research that Blobel presented, what he called the Signal Hypothesis was "imaginative," said Cook, but not well supported. "Many were unconvinced," according to Cook. "Nevertheless, the Signal Hypothesis gained gradual acceptance as a ton of data from many laboratories supported and extended it." And, wrote Cook, "(Blobel) has several times cheerfully acknowledged that he launched his idea at that Biology Division conference."

1999 Nobel Prize in Medicine Dr. Guenter Blobel, a German-born Biologist and at present working at the Rockefeller University in New York, won the 1999 Nobel Prize in Medicine.
Guenter Blobel in his laboratory The brief message delivered by the Karolinska Institutet on October 11, stated that: "The Nobel Assembly has today decided to Award the 1999 Nobel Prize in Physiology or Medicine to Gunter Blobel for the discovery that proteins have intrinsic signals that govern their transport and localization in the cell".
This apparently simple statement contains the essence of the working mechanism of the cell.
Since 1970, Dr. Blobel has been investigating a process that puzzled scientists: how proteins manage to find within the cell their proper membranes and compartments, and how do these relatively large molecules traverse the compartmental membranes without interfering with the essential gradients that exist across each of these membranes.
In 1967, Dr. Blobel began to work at the famous cell biology laboratory of George Palade a pioneer in this field who won a Nobel Prize in 1974 working at his Rockefeller Institute. In fact, Dr. Blobel's research is a direct extension of the work Dr. Palade started years before.

How do proteins translocate into the lumen of the rough endoplasmic reticulum?
What is different about the protein that is destined for the rough endoplasmic reticulum? [Note: This section describes work that led to a Nobel Prize in Medicine and Physiology to Dr. Gunter Blobel. For more information about Dr. Blobel's work and the pioneering discoveries, click: http://www.nobel.se/medicine/laureates/1999/ The major difference is the fact that it has a hydrophobic signal sequence. This simplified cartoon shows that this is the first part of the protein produced. After the signal sequence is completed, protein synthesis is further inhibited. This is to allow the interaction of the signal sequence with a complex on the rough endoplasmic reticulum. In the above cartoon, note that the signal peptide is allowed to enter and essentially guide the protein into the lumen of the rough endoplasmic reticulum. Once the signal sequence is detected, protein synthesis resumes and the rest of the protein is inserted in the lumen. Note that a signal peptidase near the inner surface of the membrane works to cleave the signal sequence from the growing peptide. The text reading for this discussion is Alberts et al, Molecular Biology of the Cell, third edition, Garland Publishing, 1994, pp 577-588 (Chapter 12) and pp 599-616.

Summary
A large number of proteins carrying out essential functions are constantly being made within our cells. These proteins have to be transported either out of the cell, or to the different compartments - the organelles - within the cell. How are newly made proteins transported across the membrane surrounding the organelles, and how are they directed to their correct location? These questions have been answered through the work of this year’s Nobel Laureate in Physiology or Medicine, Dr Günter Blobel, a cell and molecular biologist at the Rockefeller University in New York. Already at the beginning of the 1970s he discovered that newly synthesized proteins have an intrinsic signal that is essential for governing them to and across the membrane of the endoplasmic reticulum, one of the cell’s organelles. During the next twenty years Blobel
