The structural basis for the regulatory differences between the plant and fungal alternative oxidases are being assessed by carrying out a series of domain swapping studies on E. coli -expressed plant and fungal proteins. In addition, mass spectrometry is being applied to the study of the regulation of electron flow through the alternative oxidase during respiration in intact plants based on the differential fractionation of the isotope O-18 by the alternative and cytochrome c oxidases. Transformed Arabidopsis having over- and underexpressed wild-type, as well as altered forms of the alternative oxidase lacking normal regulatory control have been developed and are being used to characterize the role played by the alternative oxidase in response to different developmental and environmental circumstances, including extremes of temperature, pathogen infection, and nutrient limitations.
